RESUMO

ETHNOPHARMACOLOGICAL RELEVANCE: Fructus Choerospondiatis is the dried and mature fruit of Choerospondias axillaris (Roxb.) Burtt et Hill. It has been used for a long time in Tibetan and Mongolian medicine, first recorded in the ancient Tibetan medicine book "Medicine Diagnosis of the King of the Moon" in the early 8th century. Fructus Choerospondiatis shows multiple pharmacological activities, especially in treating cardiovascular diseases. AIM OF THIS REVIEW: This paper reviews the progress in research on the botanical characteristics, traditional uses, chemical constituents, pharmacological activity, clinical studies, and quality control of Fructus Choerospondiatis. This review aims to summarize current research and provide a reference for further development and utilization of Fructus Choerospondiatis resources. METHOD: The sources for this review include the Pharmacopeia of the People's Republic of China (2020), theses, and peer-reviewed papers (in both English and Chinese). Theses and papers were downloaded from electronic databases including Web of Science, PubMed, SciFinder, Scholar, Springer, and China National Knowledge Infrastructure.The search terms used were "Choerospondias axillaris", "C. axillaris", "Choerospondias axillaris (Roxb.) Burtt et Hill", "Fructus choerospondiatis", "Guangzao", "Lapsi", and "Lupsi". RESULTS: Fructus Choerospondiatis contains polyphenols, organic acids, amino acids, fatty acids, polysaccharides, and other chemical components. These ingredients contribute to its diverse pharmacological activities such as antioxidant activity, protection against myocardial ischemia-reperfusion injury, anti-myocardial fibrosis, heart rhythm regulation, anti-tumor, liver protection, and immunity enhancement. It also affects the central nervous system, with the ability to repair damaged nerve cells. CONCLUSION: Fructus Choerospondiatis, with its various chemical compositions and pharmacological activities, is a promising medicinal resource. However, it remains under-researched, particularly in pharmacodynamic material basis and quality control. These areas require further exploration by researchers in the future.

RESUMO

Two undescribed alkaloids, along with seven known compounds, were isolated from the roots of Zea mays (RM). Their chemical structures were elucidated based on extensive analyses of HR-ESI-MS, 1D and 2D NMR, and CD spectra. Two new alkaloids exhibited moderate inhibition of Hep3B (IC50 values of 11.7±2.4 and 14.2±3.6 µM) and SW480 cells (IC50 values of 33.4±8.2 and 47.3±5.8 µM) compared to that of the positive control compound, Oxaliplatin, IC50 value of 8.4±1.7 and 45.8±5.6 µM, respectively.

Citrullus colocynthis (L.) Schrad. (Cucurbitaceae) is widely distributed in the desert areas of the world. The fruit bodies of C. colocynthis are recognized for their wide range of nutraceutical potential, as well as medicinal and pharmaceutical uses. The plant has been reported for various uses, such as asthma, bronchitis, cancer, colic, common cold, cough, diabetes, dysentery, and jaundice. The fruit has been extensively studied for its biological activities, which include insecticide, antitumor, and antidiabetic effects. Numerous bioactive compounds have been reported in its fruit bodies, such as essential oils, fatty acids, glycosides, alkaloids, and flavonoids. Of these, flavonoids or caffeic acid derivatives are the constituents associated with the inhibition of fungal or bacterial growth, whereas eudesmane sesquiterpenes or sesquiterpene lactones are most active against insects, mites, and nematodes. In this review, the scientific evidence for the biological activity of C. colocynthis against insecticide, cytotoxic, and antidiabetic effects is summarized.

Cadmium (Cd) accumulation in brown rice is a complex process in agroecosystems and is influenced by multiple factors, such as climate, soil properties, and nutrient transport. However, during the Cd transport process (soil-root-straw-brown rice), it remains unclear how Cd concentration in brown rice (BCd) is causal relationship to environmental factors and nutrient transport. The differences in precipitation, soil properties, nutrient transport, and Cd transport were studied through a three-year fixed-point field trial and linked them to the standard of Cd and nutrient absorption and transport processes. The results showed that the available Cd concentration (ACd), and BCd in 2020 were lower than those in 2019 and 2021, but monthly precipitation (MP) was higher in 2020 than in 2019 and 2021. The MP and niche metrics were significantly negatively associated with ACd and BCd. However, the relationship between the form and location of different nutrient elements and Cd in roots, Cd in straws, and BCd also varied during the transport of nutrient elements and Cd from soil to root to straw to brown rice. Structural equation modelling analysis showed that nitrogen (N 15.5 %), phosphorus (P 14.1 %), silicon (Si 4.2 %), and iron (Fe 7.6 %) transport were more closely related to BCd than to potassium (K), calcium (Ca), magnesium (Mg), and manganese (Mn). The increase in MP significantly inhibited the increase in BCd, whereas the MP led to a decrease in BCd by affecting the transport of N and Fe. Among them, Si, Fe, and BCd had indirect causal relationships, whereas N, P, and BCd had direct causal relationships. Particularly, P is a crucial nutrient in reducing BCd in the Cd transport process. Our results highlight a strong causal relationship between environmental factors and nutrient transport and BCd, and provide a theoretical basis for fertiliser application in Cd-contaminated agroecosystems.

Zea mays (Z. mays) is one of the main cereal crops in the world, and it's by-products have exhibited medicinal properties to explore. This article intends to review the chemical compositions and pharmacological activities of by-products of Z. mays (corn silks, roots, bract, stems, bran, and leaves) which support the therapeutic potential in the treatment of different diseases, with emphasis on the natural occurring compounds and detailed pharmacological developments. Based on this review, 231 natural compounds are presented. Among them, flavonoids, terpenes, phenylpropanoids, and alkaloids are the most frequently reported. The by-products of Z. mays possess diuretic effects, hepatoprotective, anti-diabetic, antioxidant, neuroprotective, anti-inflammatory, anti-cancer, plant protection activity, and other activities. This article reviewed the phytochemistry and pharmacological activities of Z. mays for comprehensive quality control and the safety and effectiveness to enhance future application.

OBJECTIVES: The conducted search reveals that Juglans mandshurica Maximowicz, called Manchuria walnuts, had many local uses that can be categorized into cancer, dermatosis and acesodyne. KEY FINDINGS: Various metabolites including diarylheptanoids, naphthoquinones, and flavonoids (also their glycosides) were reported as bioactive metabolites. The isolated metabolites and extracts from J. mandshurica showed different biological activity including cytotoxicity, anti-inflammation, antimelanotic, anticomplement, anti-HIV, antimicrobial and anti-obesity activity. SUMMARY: It is indicated that this review will add value to more scientific research on J. mandshurica and enhance the increased interest in the sustainable commercialization of J. mandshurica. It also leads to the validation of unverified ethnobotanical claims. Future studies on J. mandshurica would be focused to establish the links between the pharmacological activity, bioactive metabolites, and traditional uses.

Carbohydrate-based drug discovery has gained more and more attention during the last few decades. Resin glycoside is a kind of novel and complex glycolipids mainly distributed in plants of the family Convolvulaceae. Over the last decade, a number of natural resin glycosides and derivatives have been isolated and identified, and exhibited a broad spectrum of biological activities, such as cytotoxic, multidrug-resistant reversal on both microbial pathogens and mammalian cancer cells, antivirus, anticonvulsant, antidepressant, sedative, vasorelaxant, laxative, and α-glucosidase inhibitory effects, indicating their potential as lead compounds for drug discovery. A systematic review of the literature studies was carried out to summarize the chemistry and biological activity of resin glycosides from Convolvulaceae species, based on various data sources such as PubMed, Web of Science, Scopus, and Google scholar. The keyword "Convolvulaceae" was paired with "resin glycoside," "glycosidic acid," "glycolipid," or "oligosaccharide," and the references published between 2009 and June 2021 were covered. In this article, we comprehensively reviewed the structures of 288 natural resin glycoside and derivatives newly reported in the last decade. Moreover, we summarized the biological activities and mechanisms of action of the resin glycosides with pharmaceutical potential. Taken together, great progress has been made on the chemistry and biological activity of resin glycosides from Convolvulaceae species, however, more exploratory research is still needed, especially on the mechanism of action of the biological activities.

Cell segmentation plays a crucial role in understanding, diagnosing, and treating diseases. Despite the recent success of deep learning-based cell segmentation methods, it remains challenging to accurately segment densely packed cells in 3D cell membrane images. Existing approaches also require fine-tuning multiple manually selected hyperparameters on the new datasets. We develop a deep learning-based 3D cell segmentation pipeline, 3DCellSeg, to address these challenges. Compared to the existing methods, our approach carries the following novelties: (1) a robust two-stage pipeline, requiring only one hyperparameter; (2) a light-weight deep convolutional neural network (3DCellSegNet) to efficiently output voxel-wise masks; (3) a custom loss function (3DCellSeg Loss) to tackle the clumped cell problem; and (4) an efficient touching area-based clustering algorithm (TASCAN) to separate 3D cells from the foreground masks. Cell segmentation experiments conducted on four different cell datasets show that 3DCellSeg outperforms the baseline models on the ATAS (plant), HMS (animal), and LRP (plant) datasets with an overall accuracy of 95.6%, 76.4%, and 74.7%, respectively, while achieving an accuracy comparable to the baselines on the Ovules (plant) dataset with an overall accuracy of 82.2%. Ablation studies show that the individual improvements in accuracy is attributable to 3DCellSegNet, 3DCellSeg Loss, and TASCAN, with the 3DCellSeg demonstrating robustness across different datasets and cell shapes. Our results suggest that 3DCellSeg can serve a powerful biomedical and clinical tool, such as histo-pathological image analysis, for cancer diagnosis and grading.

RESUMO

Based on an NMR-guided method, one new monoterpenoid glycoside (1) was isolated from Anhua dark tea, together with five known compounds (2-6). The structure of the new compound was determined as 3-(5,5-dimethyltetrahydrofuranyl)-1-buten-3-ol primeveroside, and trivially named anhuaterpenoside A (1), on the basis of detailed spectroscopic analyses, and acidic hydrolysis. Compound 1 exhibits cytotoxic activity against MDA-MB-231 and SH-SY5Y cell lines with IC50 value of 23.26 µM and 18.57 µM, respectively.

OBJECTIVES: Plant-derived flavonolignans had been demonstrated to have various biological functions. They are an important class of natural products combined by a flavonoid unit and a phenylpropanoid unit. KEY FINDINGS: From the literature survey, 88 constituents from natural resources were identified. Different derivatives of flavonolignans were listed, fused phenylpropanoid unit with dioxane ring, or cyclic ether, or simple ether side chain, or lactone, and so on. Besides, the pharmacological effects of flavonolignans were summarized as well. It has a wide range of anti-tumour, antioxidant, anti-microorganic and anti-inflammatory effects. SUMMARY: This review had provided a full-scale profile of flavonolignans on its plant sources, phytochemistry and pharmacology, and also proposed some issues and perspectives which may be of concern in the future. It was greatly anticipated that the commercialization of the flavonolignans would lead to uplift the financial abilities of communities attending the growing of the flavonolignans and the relevant and potential production becoming an international herbal and pharmaceutical commodity.

ETHNOPHARMACOLOGY RELEVANCE: The Euphorbiaceae family, which contains about 300 genera and more than 5000 species, is widely distributed in different regions. Trigonostemon genus comprises a wide group of tropical and temperate plants belonging to the Euphorbiaceae family. This genus includes at least 50 species throughout tropical Asia, extending from India and Sri Lanka to New Guinea. They have been employed by local populations for the treatment of asthma, poisonous snake bites, and food poisoning. AIM OF THE REVIEW: The main aim of the review is to critically analyze the reported traditional uses, bioactive chemical constituents and pharmacological activities of Trigonostemon species. MATERIALS AND METHODS: Scientific databases, including Google Scholar, PubMed, CNKI, SpringerLink, Web of Science, Wiley Online Library and SciFinder, were searched using keywords such as "Trigonostemon", "South China", "chemical constituents", or "traditional use". Thus, available articles from 2000 to 2020 were collected and analyzed. RESULTS AND DISCUSSION: This paper provides systematic data that Trigonostemon species possess a diverse phytochemical composition, (including diterpenes, alkaloids, coumarins, lignins, sesquiterpenes, triterpenoids, flavonoids, and polyphenols) found in different plant organs. Research on Trigonostemon plants has revealed critical therapeutic properties, such as antiviral, anti-tumor, antimicrobial, anti-inflammatory, and insecticidal activities. CONCLUSIONS: It is envisaged that the current review will add value to more scientific research on Trigonostemon species and enhance/promote the increased interest in the sustainable use of Trigonostemon species as well as lead to the validation of unverified ethnobotanical claims. Future studies on Trigonostemon species would focus on establishing the links between the traditional uses, active compounds and reported pharmacological activities.

Brassica napus L. (oilseed rape) was grown with daikon and white lupin in a polyvinyl chloride split pot experiment (with no barrier between the compartments or by a nylon mesh barrier (37 µm) to license partial root interaction, or a solid barrier to stop any root interactions) to examine the effect of rhizosphere interaction on the cadmium uptake. The results showed that shoot and root biomasses of oilseed rape were 40.66% and 26.94% less than that of the monocropped treatment (solid barrier) when intercropping with daikon under the rhizosphere complete interaction. However, the intermingling of roots between oilseed rape and white lupin notably enhanced the dry biomass of oilseed rape by 40.23% and decreased with the reduction of root contact. Oilseed rape intercropping with daikon enhanced the shoot Cd concentration of oilseed rape. The shoot Cd concentration (44.8 mg/kg) of oilseed rape when intercropped white lupin under complete rhizosphere interaction were greater than those of other treatments. Additionally, the intermingling of roots played a positive role in the content of citric and malic acids when intercropping with white lupin. In all systems, the BCF values of oilseed rape >5. Therefore, intercropping with white lupin may contribute to higher biomass and increased uptake Cd by oilseed rape. We can toward sustainable positive effects on phytoremediation that based on a better understanding of rhizosphere processes.

Curcumin (CUR), a polyphenol derived from turmeric, exhibits anticancer and anti-inflammatory properties. However, it has poor water solubility, stability, and oral bioavailability. To overcome these limitations, lipid-polyester mixed nanoparticles (NPs) embedded in enteric polymer-EudragitL100-55(Eu) were formulated (CUR-NPs-Eu). NPs composed of mPEG-b-PCL have a hybrid core made up of middle chain triglyceride (MCT) and poly(ε-caprolactone) (PCL) for enhancing drug loading. The CUR-NPs with MCT content of 10% had a particle size of 121.2 ± 16.8 nm, ζ potential of -16.25 ± 1.38 mV, drug loading of 9.8%, and encapsulation efficiency of 87.4%. The transport of the CUR-NPs-Eu across Caco-2 monolayers is enhanced compared with CUR alone (1.98 ± 0.94 × 10-6 of curcumin versus 55.43 ± 6.06 × 10-6 cm/s of curcumin-loaded NPs) because of the non-disassociated nanostructure during absorption. The absolute bioavailability of CUR-NPs-Eu was 7.14%, which was drastically improved from 1.08% of the CUR suspension (CUR-Sus). Therefore, in the xenograft 4T1 tumor-bearing mice, increased drug accumulation in heart and tumor was noticed because of enhanced oral bioavailability of CUR. The chemosensitizing effect of CUR was attributed to its NF-κB reduction effect (148 ± 11.83 of DOX alone versus 104 ± 8.71 of combined therapy, ng/g tissue). The cardioprotective effect of CUR was associated with maintenance of cardiac antioxidant enzyme activity and down-regulation of NF-κB. This study provided a partial illustration of the mechanisms of chemosensitizing and cardioprotective effects of CUR utilizing the oral availability promotion effect brought by the NPs-Eu formulation. And these results further demonstrated that the capability of this NPs-Eu system in oral delivery of poorly soluble and poorly permeable drugs.

RESUMO

Two new indole alkaloids, Bufotenidine B (2) and Bufocarboline A (6), along with seven known indole alkaloids (1, 3-5, and 7-9) and three organic acids (10-12), were isolated from the water extract of toad venom. The structures of the new alkaloids were elucidated by extensive spectroscopic methods. The absolute configurations of 4, 6, and 8 were determined for the first time by electronic circular dichroism (ECD) calculations. The cytotoxic activity of all compounds was tested against human malignant melanoma cells A375 by the MTT method, and no antitumor activity was observed.

The mechanisms of intercropping increasing plant biomass, cadmium accumulation, and organic acids secreted in rhizosphere soil are still unclear. Oilseed rape and intercrops were grown in boxes separated either with no barrier between the compartments or by a nylon mesh barrier (37 µm) to license partial root interaction, or a solid barrier to stop any root interactions. Two intercropping systems (oilseed rape-faba bean and oilseed rape-ryegrass) were carried out in soil with Cd content of 5 mg/kg. The intermingling of roots between oilseed rape and faba bean enhanced the biomass of oilseed rape. However, the biomass was negatively affected implying the higher nutrient apportionment to the ryegrass than oilseed rape. Oilseed rape intercropping with both faba bean and ryegrass played a positive role in the shoot Cd concentration of oilseed rape. The intermingling of roots played a positive role in the citric and malic acids when intercropping with faba bean. A remarkable increase in water-soluble Cd and DTPA-Cd content was observed during oilseed rape-faba bean complete root interaction treatment, up to 175.00% and 46.65%, respectively, which compare with the monoculture treatment. In both systems, the translocation factor values were higher for oilseed rape (O-F system) than for the other test plants and were always >1. Thus the Cd removal potential of oilseed rape can be further improved in the future by optimizing agronomic practices and intercropping with faba bean.

Hydroponic experiments with different cadmium (Cd) concentrations were conducted to explore the impacts of nutrient allocation on the Cd enrichment of Bermuda grass. The results demonstrated with the Cd concentration increase, the accumulation of nitrogen (N), phosphorus (P) and potassium (K) in roots, stems and leaves increased first and then decreased, while the accumulation of Cd had no significant difference. The biomass and N, P and K accumulation of CD1 and CD2 were significantly different from those of CD3 and CD4, but there was no significant difference in Cd accumulation. The root N, P and K distribution ratio of CD4 increased by 47.9%, 114.3% and 64.3% compared with those of CD2 treatment, the values of stem decreased by 29.4%, 22.4% and 17.2%, and the values of leaves increased by 15.8%, 19.8% and 23.6% respectively. The K ratio of root and leaf increased and that of stem decreased. Cd reduced N and K distribution ratio of stem and increased N and K distribution ratio of root and leaf. Pearson correlation analysis showed that the accumulation of N, P and K in stems was positively correlated with the accumulation of Cd in stems, and the accumulation of N, P and K in roots, stems and leaves were positively correlated with the Cd accumulation in leaves. Bermuda grass can schedule the nutrient allocation to adapt to the Cd absorption and enrichment in different organs under different Cd concentrations. In conclusion, nutrient allocation might affect the Cd accumulation of Bermuda grass.

RESUMO

One new α-ditetralonyl glucoside (1), was isolated from the green walnut husk of Juglans mandshurica (Juglandaceae), together with twelve known compounds (2-13). The structure of the new compound was determined as (2R,4S,10S,12S)-2-[7-(12,13,16-trihydroxy-α-tetralonyl-13-O-ß-D-glucopyranoside)]- 4,8-dihydroxy-α-tetralone-4-O-ß-D-glucopyranoside (1), on the basis of detailed spectroscopic analyses, and acidic hydrolysis. Compounds 6, 7 and 11 were isolated from the genus Juglans for the first time. Compound 1-13 showed weak cytotoxic against A549 and HeLa cell lines.

The physico-chemical characteristics of N fertilizers remain poorly understood with respect to their use with rape (Brassica napus L.) to remediate Cd-contaminated soil. In this work, eight types of fertilizer (comprising physico-chemical alkaline, neutral, and acidic N fertilizers) were employed to assess the effect of soil remediation via rape at different levels of Cd contamination (0, 5, and 10 mg kg-1 Cd). The results show that the pH of rhizosphere soils was significantly higher under physico-chemical alkaline N fertilizer treatments than under physico-chemical acidic and neutral N fertilizer treatments. The physico-chemical characteristics of N fertilizers affected the rhizosphere soil pH and promoted Cd phytoextraction and accumulation by rape. In the 5 mg kg-1 Cd-contaminated soil, the Cd accumulation and bioconcentration factor value in the shoots and the Cd translocation factor value were highest with the addition of NH4Cl, a physico-chemical acidic N fertilizer. Among the physico-chemical alkaline N fertilizers, Ca(NO3)2 enabled the highest accumulation of Cd in rape shoots when soil was contaminated with 10 mg kg-1 Cd. Thus, administering physico-chemical acidic N fertilizer to soils with lower Cd concentrations provides better remediation effects by rape, whereas physico-chemical alkaline N fertilizers are more effective in soils with higher Cd concentrations. These results show that physico-chemical N fertilizers can be employed to enhance the remediation of Cd-contaminated soil by rape and simultaneously improve the yield of this crop, with implications for environmental health and sustainable agricultural development.

Based on the potential therapeutic value in targeting metabolism for the treatment of cancer, an organic arsenical PDT-BIPA was fabricated, which exerted selective anti-cancer activity in vitro and in vivo via targeting lactate dehydrogenase A (LDHA) to remodel the metabolic pathway. In details, the precursor PDT-BIPA directly inhibited the function of LDHA and converted the glycolysis to oxidative phosphorylation causing ROS burst and mitochondrial dysfunction. PDT-BIPA also altered several gene expression, such as HIF-1α and C-myc, to support the metabolic remodeling. All these changes lead to caspase family-dependent cell apoptosis in vivo and in vitro without obvious side effect. Our results provided this organic arsenical precursor as a promising anticancer candidate and suggested metabolism as a target for cancer therapies.

Anhua dark tea known as the earliest produced Chinese dark tea, has been commercially available and famous for its unique flavor and health care effect. NMDA receptors are glutamate-coupled ion channels that critically involved in survival of neuronal cells and neurodegenerative diseases. Thus, it is considered a promising target for the therapy of neurodegenerative disease. In this study, four catechins including two new catechins derivatives (1-2), together with thirteen known flavonoids were isolated from Anhua dark tea. The structures of compounds 1-2 [2S,3R-6-methoxycarbonylgallocatechin (1) and 2R,3R-6-methoxycarbonylgallocatechin (2)] were determined on the basis of their spectroscopic data. The preliminary bioassay indicated that compound 1 showed the best neuroprotective effects via N-methyl-d-aspartate (NMDA) receptors inhibition. Compound 1 protected SH-SY5Y cells against NMDA-induced injury and cell apoptosis via the modulation of NR2B expression, the activation of PI3K/Akt signaling and caspase-dependent pathway. The results suggested compound 1 would be a potent dietary therapy reagent for prevention of excitable brain injury.
